Abstract:Objective To explore the changes of ICAM-1 expression in tumor tissues treated by local chemotherapy under different ultrasonic irradiation combing Zhifuxian microbubbles. Methods 20 tumor bearing rabbits were randomly divided into four groups: Control group, without any treatment; CPMs group, only intratumoral injection of carboplatin microspheres; S2000 group, diagnostic ultrasound irradiation on local chemotherapy tumor tissue using Siemens S2000 4v1 probe combining microbubbles; AP-170 group, the same as S2000 group except therapeutic ultrasound irradiation using Australian AP – 170. Different intensities of ultrasound were studied in the above treatment groups. Immunohistochemistry and Western blot were performed to assess the ICAM 1 expression in tumor tissues after treatment. Results Immunohistochemical semi-quantitative analysis of ICAM-1 showed that the IOD in Control group was 11038.62±873.54, while 12953.44±771.91 in CPMs group, 17823.27±921.92 in S2000 group, and 23037.09±1175.46 in ap-170 group. Western blot showed that the expression of ICAM-1 in the Control group was 0.1851±0.0295, while 0.2543±0.0354 in CPMs group, 0.4815 + 0.0391 in S2000 group, and 0.6195±0.0452 in ap-170 group. The expression level of ICAM 1 in the four groups exhibit an ascending trend in turn. No significant difference was found between the Control group and CPMs group. Compared with AP-170 group, S2000 group was statistically different, while control group and CPMs group were significantly different. Conclusion Higher intensity of ultrasonic irradiation gives rise to higher ICAM-1 expression in the local chemotherapy tumor microenvironment. In this condition, the chemotherapy drugs released by carboplatin sustained release microspheres would positively influence the proliferation and metastasis of tumor cells.
