目的:研究超声造影在评估裸鼠胰腺肿瘤内血流灌注状态的诊断价值。方法:建立人胰腺癌裸鼠原位移植瘤模型,应用随机数字表法将40只裸鼠胰腺癌模型分成两组:对照组(n = 20)和实验组(n = 20),实验组动物接受重组人血管内皮抑素治疗,对照组动物接受安慰剂治疗。所有实验动物在处死之前均接受超声造影检查,测定各组裸鼠胰腺癌组织的峰值强度(PI)和曲线下面积(AUC);利用免疫组化测定VEGF和CD34在体内胰腺肿瘤组织中的含量;采用明胶-氧化铅血管造影法检测体内胰腺肿瘤单位体积血管密度。结果:实验组胰腺癌组织的PI和AUC均显著低于对照组胰腺癌组织(P < 0.05);实验组胰腺癌组织的血管密度显著低于对照组胰腺癌组织(P < 0.05);VEGF和CD34在实验组胰腺癌组织中的表达显著低于对照组胰腺癌组织(P < 0.05);胰腺癌组织的PI和AUC均与微血管密度呈正相关(r = 0.857, P < 0.05; r = 0.762, P < 0.05);裸鼠胰腺肿瘤的PI和AUC与单位体积血管密度均呈正相关(r = 0.896, P <0.05; r = 0. 675, P <0.05)。结论:超声造影血流参数PI和AUC可准确评估胰腺癌的血流灌注量,PI和AUC与胰腺癌血管密度密切相关。 |

Correlation between contrast-enhanced ultrasound perfusion parameters and blood perfusion status in nude mice pancreatic cancer model |

Abstract: |
AIM: To investigated the diagnostic value of contrast-enhanced ultrasonography in analyzing the blood perfusion status of nude mice pancreatic cancer model. METHODS: Pancreatic cancer orthotopic xenograft tumor model was established. 40 cases of nude mice pancreatic cancer model were stochastically separated into control group (n = 20) and experimental group (n = 20). The experimental group was treated with recombinant human endostatin. The control group was treated with placebo. All animals were underwent contrast-enhanced ultrasound before execution. The peak intensity (PI) and the area under the curve (AUC) were measured by the time intensity curve. Immunohistochemistry was employed to analyze the levels of VEGF and CD34 in pancreatic cancer specimens. Tumor vascular imaging was conducted using the lead oxide-gelatin injection technique. RESULTS: A significant decrease of PI and AUC were seen in experimental group specimens as compared to the control group tissues (P < 0.05). The vascular density was markedly lower in experimental group than that in control group (P < 0.05). The relative levels of VEGF and CD34 were significantly down-regulated in experimental group specimens in comparison with control group tissues (P < 0.05). The PI and AUC were positively correlated with micro vascular density in pancreatic cancer tissues (r = 0.857, P < 0.05; r = 0.762, P < 0.05). The PI and AUC were positively correlated with vascular density per unit volume in pancreatic cancer tissues (r = 0.896, P < 0.05; r = 0.675, P < 0.05). CONCLUSIONS: Contrast-enhanced ultrasound can accurately evaluate the blood perfusion of pancreatic cancer, and the values of PI and AUC are positively related to the vascular density of pancreatic cancer. |
