目的 探讨低频低强度超声辐照微泡(LILFU-MB)对MDA-MB-231/DOX细胞耐药性的影响。方法 筛选LILFU-MB非毒性照射时间;选取不同浓度(10-2μM,10-1μM,100μM,101μM,102μM,103μM)阿霉素(DOX)分别处理细胞24h,测定DOX对耐药细胞的半抑制浓度(IC50)。将细胞分为对照组、DOX组、LILFU-MB组、DOX+LILFU-MB组,检测细胞增殖、迁移、凋亡情况以及P-gp、Bcl-2、Cyt-c和Caspase-3蛋白表达水平。结果 LILFU-MB非毒性照射时间为30s,能降低DOX对耐药细胞的IC50。与单独DOX处理和LILFU-MB处理相比,DOX+LILFU-MB组细胞增殖率和迁移率最低,细胞凋亡核型改变最明显,细胞凋亡率、Cyt-c和Caspase-3蛋白表达水平均升高(P<0.05),Bcl-2蛋白表达水平显著降低(P<0.05),同时DOX+LILFU-MB组明显下调P-gp蛋白的表达(P<0.05)。结论 LILFU-MB通过下调P-gp蛋白表达,抑制MDA-MB-231/DOX细胞增殖、迁移并促进细胞凋亡,进而逆转三阴性乳腺癌MDA-MB-231/DOX细胞耐药性。 |

Objective To investigate the effect of low-frequency low-intensity ultrasound irradiation microbubbles on drug resistance in triple-negative breast cancer MDA-MB-231/DOX cells. Methods The optimal irradiation time of LILFU-MB was optimized. The MDA-MB-231/DOX cells were exposed to various concentrations of Doxorubicin (DOX) for 24h (ranging from 10-2μM to 103μM) to assess the half-inhibitory concentration (IC50) of DOX. Four groups were formed: control group, DOX group, LILFU-MB group and DOX+LILFU-MB group. Cell proliferation, migration, apoptosis, and protein expression levels of P-gp, Bcl-2, Cyt-c and Caspase-3 were evaluated. Results After a non-toxic irradiation period of 30s with LILFU-MB treatment reduced the IC50 value of DOX in drug-resistant cells. Compared to individual treatments with either DOX or LILFU-MB alone, the combination treatment in the DOX+LILFU-MB group exhibited significantly lower proliferation rate and migration rate as well as more pronounced apoptotic karyotype changes. Furthermore, this combined treatment resulted in increased apoptosis rate , elevated protein expression levels of Cyt-c and Caspase-3 (P<0.05) and decreased protein expression levels of Bcl-2 (P<0.05). Importantly, the expression level of P-gp protein was significantly down-regulated by the DOX+LILFU-MB group (P<0.05). |
